
Maheul PLOTON
Dr. Maheul PLOTON is a statutory engineer at INSERM. Since 2022, he has been the VVEDIT platform scientific and technical manager of the PRISM laboratory (INSERM U1192) directed by Prof. Michel SALZET.
His career began in 2004 when he joined unit U545 directed by Prof. Jean-Charles FRUCHART, he then continued in unit U1011 created in continuity by Prof. Bart STAELS. Within these units, Maheul developed his skills in the field of biochemistry, cell biology, bioinformatics and molecular biology. In 2008, he became the manager of a technical platform dedicated to the viral vectors production and use, taking charge of the technical and regulatory aspects but also the new tools development. This activity allows him to acquire solid expertise in the field of transgenesis using viral vectors. He participates in 12 publications, 7 of which are directly associated with this activity.
At the same time, Maheul has successively led two research projects, in the context of understanding the type 2 diabetes progression, which will result in publications for which he is first author. These works objectives were to determine the nuclear receptors action in pancreatic failure, “The nuclear orphan receptor Nur77 is a lipotoxicity sensor regulating glucose-induced insulin secretion in pancreatic β-cells. Mol Endocrinol. 2012”, and in the glucose production regulation by the liver “The nuclear bile acid receptor FXR is a PKA and FOXA2-sensitive activator of fasting hepatic gluconeogenesis. J Hepatol. 2018”. This last study, carried out as part of a thesis, allowed Maheul to obtain his PhD at the University of Lille.
Within the PRISM laboratory, Maheul manages the VVEDIT platform “Viral Vectors for genome EDiting and Transgenesis” on a daily basis. Viral vectors are cargo elements that allow the interest gene sequences transport and stable, transient or controlled expression within cells (in vitro, ex-vivo or in vivo). Maheul ensures the defective viral vectors custom development, optimization and production (bioengineering of promoters, gene sequences and viral tropisms, modified and optimized in order to the interest cells best target). In addition to these transversal activities, his research focuses on secondary resistance mechanisms involving tumor ECM in breast cancer and on a macrophage bioengineering method development in order to target cancer cells in glioblastoma.
Maheul is also involved in training. Beyond that dedicated to the viral vectors use, in connection with the platform, which he was able to provide to all types of personnel. He supervised two Master’s interns and technically trained many doctoral students in the various skills of which he acquired expertise.
Maheul is also a trainer within the DU “Conception en expérimentation animale (Design in animal experimentation) “. Teaching within which he carries out a course whose objectives are to explain the defective viral vector concept, to inform on the different viral vectors characteristics, to highlight their interests in in vivo transgenesis and to detail the methodological and regulatory steps of their implementation.
